Been living downtown for 2+ years. I'll list my suggestions by category. I can offer greater detail, if you like.

Can't remember name, but there's an authentic Italian guy in the market whose open 'til 2am on weekends. I'll be there tonight kinda late.
And wherever you go for dinner, you absolutely MUST go to Kaminsky's in the market for dessert. Unbelivable cakes, pies, and shakes.
Overall, my top 5 must try's in a great food town would be these:
1. Poogan's Porch - took wife there on night of my engagement; try the she crab soup or pimento cheese fritters to start, then have either the Plantation Chicken or Lowcountry pasta. Located near Meeting and Queen)
2. 82 Queen - neat slightly formal and very Charlestonian atosphere, great food. Located near King and Queen.
3. Fleet Landing. Decent mix of seafood and American, but by far the best view of harbor anywhere. Its been mild, so ask to sit outside.
4. Hank's - best seafood in Charleston, no matter what Hyman's claims. This is where locals go for the best. Located just of South Market.
5. Jim 'N' Nick's. Best BBQ. Lots of smoked meats. Very fuilling at reasonable prices. Tv's all around. Check out the 1920's pin-up model above the bar!
